In 1964 a reluctant John Lennon is persuaded by manager Brian Epstein to meet Freddie, the father who abandoned him 17 years earlier, with the press in attendance. The meeting is short and bitter. Three years later Epstein is dead and John invites Freddie to his mansion but again things turn sour,due to Freddie's drinking and insulting Mimi,the aunt who raised John. The Beatles set up Apple records but the press are hostile and Lennon's comment that 'we're more popular than Jesus' doesn't help. Rows with long-suffering wife Cynthia lead to marital breakdown and John's meeting Japanese performance artist Yoko Ono. Family history is repeated as Lennon leaves Cynthia and their son Julian for Ono, by whom he has a second son,Sean. In 1969 John returns his M.B.E. in protest at England's support for the Vietnam War while his stunts with Yoko Ono to promote peace alienate the press. Some months...

Maka, Peter alias Spiderman kemudian berubah menjadi agresif dan percaya diri. Bahkan cenderung jahat dalam menghadapi segala persoalan yang dihadapi. Termasuk menghadapi Harry Osborn (James Franco), sahabat Peter yang sekaligus anak dari musuh besarnya, Green Goblin, yang telah meninggal.

Harry (diakhir film Spiderman 2) mengetahui identitas Spiderman sebenarnya. Sejak saat itu bertekad untuk menuntut balas kepada Spiderman yang diduga telah membunuh ayahnya.

Selain Harry, Peter juga harus menghadapi Erick Brock (Topher Grace) fotografer freelance yang mengincar pekerjaannya di harian The Bulge.

Spiderman juga harus melindungi kota New York dari serbuan penjahat Sand Man (Manusia Pasir), Flint Marko (Thomas Haden Church). Sand dapat berubah bentuk menjadi raksasa, seperti Hulk dari pasir, tapi kuat dan tidak dapat dihancurkan lantaran dapat kembali membentuk dirinya.

Dan, seakan-akan komplikasi itu tidak cukup, sang jagoan Spiderman juga harus menghadapi masalah yang timbul dari hubungan cintanya. Karir Mary Jane sebagai pemain teater hancur dan membutuhkan perhatian Peter Parker maupun sosoknya sebagai Spiderman.

Spiderman yang sedang dirasuki mahkluk angkasa luar yang membuatnya jadi agresif dan sibuk mengejar Sand Man yang membunuh Ben (Cliff Robertson). Tidak ada waktu untuk Mary Jane yang sedang merajuk mencari perhatian.

Sisi gelap sang Spiderman itu ditampilkan dengan berubahnya kostum laba-laba merah biru menjadi hitam setelah makhluk dari angkasa luar itu merasukinya.
